Biochimie neuroendocrinienne

Notre recherche se concentre sur une famille de neuf protéines convertases (PC) de mammifères : PC1, PC2, furine, PC4, PC5, PACE4, PC7, SKI-1 et PCSK9. Ces enzymes sont impliquées dans l’activation de précurseurs d’hormones, de facteurs de croissance ou de transcription, de récepteurs, ou de protéines d’enveloppe chez les agents pathogènes. L’équipe se concentre sur le mécanisme d'activation et la régulation de ces enzymes, ainsi que l’identification de leurs substrats et inhibiteurs potentiels. Nos déchiffrons aussi en profondeur les rôles physiologiques in vivo des PCs dans les cellules, chez la souris et chez l’humain.

Notre groupe vise l’exploration des implications cliniques des proprotéines convertases dans l’état pathologique, comme pour la PCSK9 dans l’hypercholestérolémie. Notre équipe utilise une vaste gamme de techniques : biochimie des protéines et de peptides, enzymologie, biologie moléculaire et cellulaire, protéomique, génétique. 

Le laboratoire possède une collection unique de PCs mutantes et a aussi des souris knockouts (complet ou conditionnel) pour PC5, furine, PC7 et PCSK9. La PCSK9 est une cible particulière pour le traitement de l’hypercholestérolémie familiale. Des traitements anti-PCSK9 sont maintenant disponibles mondialement. Nous avons aussi établi un nouveau rôle de l’inhibition de PCSK9 dans le traitement de cancers/métastases et de certaines infections virales. Nous avons récemment découvert un nouveau rôle pour PC7 dans la régulation des triglycérides et la stéatose hépatique, et nous développons des approches de silençage par oligonucléotides antisens afin de traiter cette pathologie. Les rôles clés que jouent SKI-1 et PCSK9 dans l’homéostasie du cholestérol ont mené l’équipe à élargir ses horizons et à consacrer des efforts considérables dans le développement d’applications pharmacologiques reliées aux maladies cardiovasculaires et cardiométaboliques.

Enfin, nous avons récemment démontré le rôle critique des convertases qui s’apparentent à la furine, dans l’activation de l’entrée en cellule de SARS-CoV-2, l’agent éthologique de la COVID-19. De petites molécules inhibitrices sont maintenant offertes pour des études précliniques, comme antiviraux en anticipation de futures pandémies.

Profilage fonctionnel 

Chaque convertases et leurs inhibiteurs/silenceurs sont testés dans des cellules et dans des modèles animaux, puis finalement chez des patients humains.

Biochimie et génomique

Le mécanisme d’action de chacune des convertases et l’identification de substrats physiologiques et pathologiques et/ou cibles dans les états pathologiques.

Modélisation des maladies

Utilisation des souris KO et transgènes ainsi que les SNPs humains et variants comme indicateurs de l’importance fonctionnelle de chacune des convertases et des traitements potentiels en milieu clinique.
