We are looking for a talented research assistant to participate in the development of the RNA Therapeutics Core Facility at the Sidney-Altman Hub at the Montreal Clinical Research Institute (IRCM).
You will play a central role in establishing the core facility services and in the execution of activities. This position offers a unique opportunity to contribute to cutting-edge research in RNA therapies and to stimulate innovation in RNA biology.
Summary of responsibilities
Project management and execution:
- Perform research design and planning of RNA therapeutic projects together with the core facility manager.
- Carry out laboratory work, including:
- Synthesis, purification and NGS of sc-pDNA plasmids and vectors
- Design of RNA sequences and vectors
- In vitro RNA synthesis, purification, translation, and associated quality control assays using HPLC, Nandrop, ELISA, Flow Cytometry, etc.
- Analysis of qualityof DNA templates such as Poly A tail, linearization efficiency, and quantification.
- Quality control and analysis of RNA using Nanodrop, ELISA, nanopore sequencing
- Formulation of lipid nanoparticles and RNA encapsulation, and quality control monitoring
- Screening in cell culture test dose-response effects
- Assist with synthesis of oligonucleotides, e.g. ASO, siRNA, sgRNA including high throughput containing standard and/or modified nucleobases, as well as purification, lyophilization, and analysis thereof.
- Apply a wide range of molecular biology techniques and core lab skills and develop and/or optimize experimental procedures, e.g., for designing, identification, quantification, and purification of plasmid DNA and PCR amplified templates, plasmid linearization, bioconjugation, etc.
- Create, collect and interpret experimental data with strict adherence to data integrity according to GLP guidelines.
- Assist in establishing a quality control measure to ensure the highest standards in RNA, oligonucleotide and nanolipid particle synthesis and purification processes, screening, cell culture assays, and in vivo functional assays.
Operational and collaboration:
- Contribute to the development of the service offering of the RNA therapeutic core facility with the manager.
- Support collaborative interactions with partner laboratories, institutions and clients.
Required qualifications:
- Master’s degree (MSc) in molecular biology, biochemistry, bioengineering or a related field.
- Two-three years industry experience.
- Experience and skills in cell culture and cell-based assays, PCR (various), in vitro RNA synthesis, HPLC, FPLC, Mass Spectrometry, Capillary Electrophoresis, Lipid nanoparticles, DLS, UF/DF, Nanodrop
- Strong background in mammalian and bacterial cell culture, recombination and transformation, broad range of molecular biology techniques such as cloning, amplification, transcription, translation, and associated assays and bioanalytical techniques including method development and qualification.
Knowledge and skills:
- Previous exposure to therapeutic RNA applications.
- Demonstrated knowledge of fundamental principles of RNA biology.
- Superior computer literacy: MS-Office and planning tools, i.e. Smart Sheets, Project, Excel (PivotTable, data trending, etc.)
- Strong analytical, problem-solving and communication skills.
- Excellent organizational and documentation skills.
- Strong and the ability to multitask and prioritize effectively.
- General knowledge of chemical synthesis of oligonucleotide and ligation.
- Understanding of cGMP (Good Manufacturing Practice) requirements.
- Exposure to LIMS and QMS.
