Abstract:
I work at the interface of cell biology, cancer biology, and mechanobiology to define the factors that influence how tumors interact with neighbouring tissues as they grow and invade. To this end, I will discuss my work applying spatial and single-cell transcriptomics and quantitative microscopy to a zebrafish model of melanoma. This approach revealed that cells spanning the tumor border are reprogrammed to support tumor invasion, and uncovered a novel role for cilia in invasion. This spatially-restricted reprogramming is mediated by the chromatin modifier HMGB2, which is highly upregulated at the tumor border. I will describe new work uncovering a novel mechanosensitive role for HMGB2 in invasive tumor cells, modulating chromatin architecture, gene expression, and cell state in response to mechanical force. Finally, I will discuss my plans for future work focused on the influence of the mechanical microenvironment on tumor cell biology and tumor progression.
