About this conference
The Mediator complex is essential and conserved coactivator that plays a crucial role in transcription regulation across eukaryotes. A growing body of evidence suggests that Mediator complex plays more roles in nuclear processes, serving as a link between transcription, genome organization and DNA repair including nucleotide excision repair. Thus, we revealed that Mediator cooperates with RSC, an essential chromatin remodeler, in chromatin organisation and transcription. Moreover, we propose that Mediator acts in transcription-coupled repair for recruitment of DNA repair machinery. Mutations in human Mediator are implicated in many diseases, including cancers or neurodevelopmental diseases recently regrouped as MEDopathies. However, in many cases, the underlying molecular mechanisms remain to be understood. We are addressing these questions by using the yeast Saccharomyces cerevisiae as our main model, and by applying functional genomics, genetics, microfluidics, bioinformatics and modelling.
